1. Pipeline: (3D) Lipid Droplet in Adipocyte
This pipeline is designed to segment lipid droplets (LDs) in adipocytes, where droplets range widely in size from small to very large. To accurately capture the full LD population, it combines refractive index (RI) thresholding with a tophat-based post-processing technique. This hybrid approach enhances detection sensitivity, enabling robust segmentation of both low-contrast small droplets and large, dominant LDs within the same cell.
The pipeline provides comprehensive LD quantification across the entire adipocyte volume, supporting detailed analysis of lipid storage and distribution in 3D.
Output: Labeled individual lipid droplets, LD measurements

2. (2D) Lipid Droplet in Adipocyte
This pipeline is tailored for analyzing lipid droplets (LDs) in adipocytes, which often exhibit a wide size range from small, low-contrast droplets to large, dominant ones. To capture this diversity accurately, the pipeline combines refractive index (RI) thresholding with a tophat-based post-processing method. This rule-based approach enhances the segmentation of both subtle and prominent LDs in 2D projections.
The pipeline enables reliable quantification of lipid droplet number, area, and size distribution, making it suitable for studying lipid accumulation, metabolic activity, and adipocyte differentiation using label-free holotomographic imaging.
