Dynamic Live Cell Imaging in Microfluidic Chips Using Low-Coherence Holotomography

thum_cellBio-poster_01.png

Applies the all-in-one HT-X1 holotomography system, with its integrated incubation stage, to observe live-cell dynamics label-free inside three commercial microfluidic devices (µFDs) — examining subcellular resolution, collagen-matrix effects on cell morphology, and cellular response to oxidative stress under perfusion.

- Subcellular-resolution HT imaging of K562 cells and mitochondria-DsRed Hep3B cells in a flat multi-channel µ-Slide, including HT–fluorescence correlation of mitochondria
- 3D collagen type I culture (DAX-1 chip) showing Hep3B cell–matrix contact and collagen fiber network structure across pH 5.0/7.4/11.0 polymerization conditions
- Perfusion-system schematic and time-lapse imaging of cellular response and recovery from 2.5 mM H₂O₂-induced oxidative stress (0–270 min)
- Discussion of HT's ability to resolve mitochondria, membrane, nucleus, and lipid droplets inside microfluidic devices without labeling
