Characterizing Mitochondrial Fission/Fusion Dynamics via Holotomography Imaging

Frame 1712.png

Demonstrates label-free, high-speed HT imaging of mitochondrial morphology in Hep3B cells, validating RI-based mitochondria identification against a fluorescent mitochondrial marker, then applying an ML-based TomoAnalysis pipeline to quantify drug-induced (Mdivi-1/MPP+) and time-dependent shifts between fragmented and elongated mitochondrial states.

- RI line-profile validation of mitochondria against Mito-DsRed fluorescence signal
- 30-second, 6-second-interval time-lapse imaging capturing individual mitochondrial fission and fusion events
- Machine-learning (pixel classification) segmentation pipeline extracting per-mitochondrion volume, surface area, dry mass, and length
- Dose-dependent (Mdivi-1 fusion inducer / MPP+ fission inducer) and time-dependent (0–32 min) stacked bar-chart quantification of fragmented/normal/elongated mitochondria populations
