Observation and AI Supported Analysis of Lipid Droplet Dynamics in Live Organoid Using Label-Free 4D High-Resolution Holotomography

Uses an 84-hour, 96-well-plate holotomography screening workflow to induce and quantify a hepatosteatosis model in mouse hepatic organoids (mHO) via free-fatty-acid treatment, combining rule-based and AI-based (ilastik) segmentation to track lipid droplet accumulation and cell-layer dry mass changes.

- HT-Ready 96-well-plate screening design testing oleic acid, palmitic acid, linoleic acid, and combinations at multiple concentrations over 84 hours
- Time-lapse (0–84 h) label-free LD accumulation imaging across all treatment groups, with zoomed single-organoid detail
- Combined rule-based + AI-based LD segmentation pipeline with 3D renderings at 0 h and 60 h
- Quantitative LD count and volume trends per treatment, plus cell-layer/lumen segmentation pipeline showing dry-mass decline in lipotoxic groups vs. increase in other groups
